At the end of Chapter 355, it says on the bottom of the page that Negima is diverting between the main story line and a movie that has "Ending B" written by Akamatsu. Discuss?

Nothing much, just an alternate ending to something (maybe this chapters end) that he wrote as a treat to us.

Just a different path between anime and manga. The OVA will be the alternate story after the magical arc, diverging from the original story planned in the manga.

Would I have to have watched the Negima anime in order to enjoy the movie? I hope not because I feel like watching it, but don't have the patience to watch the series. I wonder how this "Ending B" will differ from the actual ending? I wouldn't mind a ten year skip since it would show us what the future is like for Negi and his students.

The 'movie' was apparently extremely dumbed down and SHAFT didn't have enough time in their planned production schedule and could not finish the movie in time properly, resulting in unfinished, unpolished movie that would warrant either addition of scenes or secondary ending.
On the opening celebration, one person apparently said "In terms of entertainment, it is ordered like this: the Greetings by the Casts -> Hayate -> Negima. I'd say it's pretty understandable seeing how there was like a scene with five full minutes of white screen with only explosion sound effects.
