I think everyone and anyone is understanding when you need a more challenging and satisfying job than the one you currently have. In the end it is your daily life it is your time you spend there, not theirs.
In other words, if he is a relative that you know good enough, he'll accept it. If he doesn't accept it, he most likely doesn't care about your future either.
Regarding the letting him down part. Just tell him early on that in the next "x" weeks you are going to switch jobs / going to look for a new one so that it won't come out of the blue for him. That way you can give him time to find a replacement and take your own time to find a new job, incase you don't have another offer already.
Yup , I've done the exact thing that you mentioned , telling him that next month I'm going to move outta town seeking for a new job. Then his expression went pale as if it's the end of the world , perhaps he never thought about me -his close relative- to leave him this sudden.
Well watching his grave expression is enough to drain the colour out of my face.

Gahaha I would never take it on the wife , she isn't always a bitch , she could be an angel in 1 out of 10 occasions , well the problem is those other 9 occasions.....
The job is quite decent actually , it's just the boredom I couldn't take , and the fact that I can't open FAKKU while I'm at work !!
