Anime tends to be a little too superficial to have any lasting affection for characters, imo. VN tend to be a better place for that. If I need pick a character from anime, probably Ushio from Clannad, or Senjougahara Hitagi from Bakemonogatari for very different reasons. Maybe Momoko from Sumomomo Momomo.
Were I to pick a VN girl, definitely Mikan from Wanko to Kurasou, with special mention of Sakura from F/SN and Sora from Ever17.

I guess the first for me would have to be Ogiue from Genshiken, to the point where I was seriously annoyed that she wasn't real. After that it was Taiga of course, and then pretty much just Tsundere in general. It's too bad they just come off as bitchy IRL. :(
