Not only TLC all kind of chromatographies. I have exam in proteins and how to purifying them and analysing them based on size or pH etc.

Filling up a column:
Top: Sand
Silica (lots of it)
Sand
Bottom: Cotton to prevent sand and silica from getting out.

The fundamental principle when a column runs is the differing polarities of the compounds being filtered. Because of this, different compounds have different speeds as they move through the silica, causing them to separate from other compounds and clump together
