That's a hard question to answer. All of them have their faults. Time on the site, for instance, can be misleading. I signed up in December of 2007, but I almost never visited the forums until this past June or July. As for post count, all it takes to get a high post count is MoteMote. Show up when that starts, and post in all the threads, and within a month, you'll be an elder. People probably won't like you though. And to get rep, all you really need to do is post stuff in User Uploads. Well, that's the fastest way to get a lot of rep.
I think it's a combination of all of those, time on the site, post count, and rep. Having just one doesn't mean a whole lot, but it's hard to bullshit past all three.
But ultimately, doesn't the user him/herself define what makes a person a senior user? That is to say, the person who is looking at the user creates his/her own definition of what makes a person a senior user. If new user Billy Buttfuck thinks that rep is all that matters, then that's what he's going to look at, and if Amanda Hugginkiss thinks post count is the most important thing, then she's going to think the people with the highest post counts are the best. After all, most of the regular users know who we respect because of what we've seen, not because of statistics.
