They can't. It's mentioned in the visual novel. I think Illya specifically mentions that during the Heaven's Feel scenario IIRC.
IIRC, any assassin was originally infamous. But I think you can do that for any other class as well, so long as you have an item connected to them.

False. The Assassin class was implemented at the start by a rule; Assassins would always come from one of the 19 Hassans. Personality/actions didn't matter; Assassins would come from one of the Hassans. The fake Assassin (Sasaki) during the 4th HGW was an error allowed by the corrupted Holy Grail. Pretty sure that was mentioned in the visual novel.
